FAQ

At Resea Biotec, we understand that you may need immediate help. That’s why we’ve put together this collection of frequently asked questions to assist with the maintenance of Applikon products.

These common topics may help you when handling your Applikon controller and bioreactor. We aim to keep these FAQs updated. If there is an issue you think we should add to the list, please let us know!

The heater tank of a Thermo Circulator contains a float that is the water detector. In the case of an empty heater tank, an error message will be presented and the heating element will be disabled.

The heater tank must be filled with water before the actuator can be put to use. This screen can be opened using the following navigation path:

Home>Menu>Configuration>Actuators>TC

Press the “fill” button in the Additional Configuration section of the screen. As soon as the Thermo Circulator is filled with water, press the “stop filling” button. The press the “activate” button.

Note: you will know when the Thermo Circulator is filled with water when the warning sign in the “Menu” button disappears and water flows into the drain.

The Stirrer Speed Controller (a control loop itself) can be used as an actuator that is assigned to the dO2 Control Loop (an increasing stirrer speed will result in better dispersion of the gas bubbles in the medium). In that case, the lower and upper limit of the stirrer speed can be set using the so-called “Cascade Limits”.

The Cascade Limits are found in the Stirrer Settings > Limits screen (indicated by a red rectangle in the adjacent image).

When the stirrer speed controller is used as a (manual) stand-alone control loop, the Cascade Limits are disabled.

When mounting the head plate on top of the reactor, ensure you fix the mill nuts / slotted nuts in a crosswise manner! Please don’t use any tools to fix the screws of the bioreactor – your WoMan power is sufficient!

  • P-Gain:
    • Proportional gain; causes a controller output that is proportional to the deviation
  • I-interval:
    • Integrates the deviation to correct for a systematic offset
  • D-interval:
    • Evaluates the trend and prevent a large overshoot
  • Dead Band
    • Prevents excessive actuator actions when the setpoint is reached
  • Cycle Time
    • Defines the Control Loop refresh time

During the cultivation, the Sample Correction procedure corrects the online measured pH value with data from offline pH measurements in samples. Only the current offset is corrected by entering the off-line measured value.

Press the “sample correction” button and use the numeric keypad to enter the pH value measured in the sample. The measured value will be corrected and the difference between the online value and the corrected value is displayed on the calibration screen.

The control software is able to detect a “Low Gas Flow” occurrence and will generate an alarm.

  • When the actual measured gas flow is smaller than 50% of the required gas flow, the Low Flow alarm is triggered. In an attempt to cure the problem, the software will retry to open the block valve (max. 5 times).
  • If the issue is not resolved automatically, please get in contact with us. Often, the gas inlet flow is to low. the magnetic valve doesn’t open properly or the valve is not properly aligned with the MFC.

The dO2 measurement is based on the polarographic principle (Clark-cell). Therefore, the sensor must be polarized for at least 4 hours before it can be calibrated. If the sensor is calibrated before polarizing, the measurement value can differ from the actual value in the vessel.
